A new substrate for protein tyrosine phosphatases (PTPases) is described. The substrate is composed of a dansyl group attached to a tyrosine residue on a protein through a cysteine residue. The dansyl group is incorporated in the PTPase's active site to form a Michael-type conjugate. The covalent attachment of the dansyl group to the tyrosine residue is thermodynamically favored over its random attachment. Thus, under basic conditions, where PTPase activity is low, the modified tyrosine is incorporated into a protein by a reaction that is faster than the random attachment of the dansyl group. However, in the presence of active PTPase, the PTPase activity causes dephosphorylation of the protein, which enhances the incorporation of the dansyl group. The substrate displays a marked fluorescence increase when incubated with various active PTPases, especially high activity PTPases.
